An antiviral nucleoside analog and a prodrug form of GS-441524; inhibits the replication of SARS-CoV-2 and SARS-CoV-2 Βeta, Gamma, or Omicron variants in infected Vero E6 cells (EC50s = 1.36, 1.127, 0.349, and 0.106 µM, respectively) without inducing cytotoxicity (CC50 = 128 µM); reduces nucleocapsid gRNA copy number and nucleocapsid sgRNA levels in the lungs of, and decreases lung damage in, a KI-hACE2 mouse model of SARS-CoV-2 infection at 250 or 500 mg/kg per day; reduces the levels of S protein in the lungs of, as well as decreases PFUs in lung tissues, reduces lung and spleen inflammation, and prevents viral infection-induced death in, a K18-hACE2 mouse model of SARS-CoV-2 infection at 100 or 250 mg/kg per day
